报告题目:Cooperative conformational changes of actin filaments allosterically regulate interactions with actin binding proteins.

报告摘要:Diverse functions of actin filaments are dependent on interactions with specific actin binding proteins (ABPs). Local biochemical regulation of each ABP has been implicated in localizing those interactions in vivo, but polymorphism of actin filaments is emerging as a novel mechanism of functional regulation of actin filaments. We present evidence to show that mutually exclusive cooperative binding of S1 (motor domain of myosin II) and cofilin to actin filaments is mediated by cooperative conformational changes of the filaments. High-speed AFM suggested that S1-mediated inhibition of cofilin binding involves cooperative untwisting of the helical filament structure. This is consistent with previous reports that stretching actin filaments inhibits cofilin binding (Hayakawa et al., 2011) and promotes S1 binding (Uyeda et al., 2011) in vivo, since MD simulation suggested that stretching actin filaments untwists the helix (Matsushita et al., 2011). Similar allosteric regulation of ABPs may be widespread since many ABPs are known to change the filament structure.
